Design Notes

To make sure the ASTERISK-RNA works as it is supposed to be, we made sure the Sensor-RNA itself is free from restriction cloning scars. To provide a working and approachable System for other teams we designed this basic part as working out of the box. For this we describe in vitro translation and transfection based on lipid nano particels.


Source

ASTERISK works by using the MCP-ADAR2DD(E488Q)-NES enzyme found in human cells. This enzyme binds to a MS2 RNA hairpin derived from Bacteriophage MS2. The example RNA described in this Part contains sequences for mCherry, mNeonGreen.
